Bistramide A is a marine natural product with a dual mode of action. It was originally identified as a specific PKC-delta activator, but recent studies by Kozmin et al. have identified actin as a principal target. We have recently completed the total synthesis of Bistramide A, and are interested in preparing a library of analogs in order to further probe the biological activity: Can we identify analogs that bind to actin, but do not activate PKC-delta? Similarly, can we identify analogs that activate PKC-delta but do not bind actin?
The synthesis is based on the preparation of spiroketals from carbohydrate derived lactones via exo-enol ethers, while the tetrahydropyran system is prepared by a kinetically controlled oxa-Michael cyclization. Different methods will be used to couple the three fragments.

L’Attenol A was isolated in 1999 from Pinna attenuata and showed cytotoxic activity against P388 Leukemia cells. A total synthesis will be performed via the corresponding exo-cyclic enol ether.

Broussenetine H and N will each be synthesized by coupling of a spiroketal fragment derived from valerolactone and a carbohydrate-derived pyrrolidine.
